چکیده

conclusions lrrfip1 and its mediated production of type i interferon play a role in controlling hcv infection. the findings of this study provide new target for hcv treatment and contribute to development of anti-hcv drugs. results lrrfip1 increased the expression of ifn-β in hepatocytes with or without hcv infection. induction of ifn-β by lrrfip1 was enhanced with the presence of hepatitis c virus. overexpression of lrrfip1 in hepatocytes inhibited hcv replication. however, hcv infection did not regulate intracellular expression of lrrfip1. materials and methods induction of ifn-β by lrrfip1 in huh7 and huh7.5.1 was determined by real-time pcr and western blotting in vitro. inhibition of hcv replication by lrrfip1 overexpression in hepatocytes was assessed. objectives the aim of this study was to assess the role of lrrfip1 in induction of ifn-β and inhibition of hcv infection in hepatocytes. background hepatitis c virus infection is one of the leading causes of end stage liver diseases. the innate immune response slows down viral replication by activating cytokines such as type i interferon (ifn-α/β), which trigger the synthesis of antiviral proteins and modulate the adaptive immune system. recently, leucine-rich repeat (in flightless i) interacting protein-1 (lrrfip1) was reported contributing to the production of interferon-β in macrophages.

LRRFIP1 Inhibits Hepatitis C Virus Replication by Inducing Type I Interferon in Hepatocytes

Recently, leucine-rich repeat (in Flightless I) interacting protein-1 (LRRFIP1) was reported...

Alpha interferon inhibits hepatitis C virus replication in primary human hepatocytes infected in vitro.

Chronic hepatitis C is a common cause of liver disease, the complications of which include cirrhosis and hepatocellular carcinoma. Treatment of chronic hepatitis C is based on the use of alpha interferon (IFN-alpha). Recently, indirect evidence based on mathematical modeling of hepatitis C virus (HCV) dynamics during human IFN-alpha therapy suggested that the major initial effect of IFN-alpha i...

Recombinant duck interferon gamma inhibits duck hepatitis B virus replication in primary hepatocytes.

Interferon gamma (IFN-gamma), which has been cloned in several mammalian species and recently in birds, plays a critical role in modulating immune system function. IFN-gamma and tumor necrosis factor alpha (TNF-alpha) have been shown to be crucial in the pathogenesis of viral hepatitis and in the transient disappearance of hepatitis B virus (HBV) from the liver after adoptive transfer of HBV-sp...

(-)-Epigallocatechin-3-gallate enhances poly I:C-induced interferon-λ1 production and inhibits hepatitis C virus replication in hepatocytes

AIM To investigate the effect of (-)-epigallocatechin-3-gallate (EGCG) on polyinosinic-polycytidylic acid (poly I:C)-triggered intracellular innate immunity against hepatitis C virus (HCV) in hepatocytes. METHODS A cell culture model of HCV infection was generated by infecting a hepatoma cell line, Huh7, with HCV JFH-1 strain (JFH-1-Huh7). Poly I:C with a high molecular weight and EGCG were u...

Comparison of PEG Interferon Loaded and non-Loaded Iron Oxide Nanoparticles on Hepatitis C Virus Replication in Cell Culture System

Background and Aims: Iron oxide nanoparticles are among the most effective tools which can replace current medical techniques for diagnosis and treatment of various diseases. Hepatitis C infection is one of the main health problems in the world, affecting around 3% of the world's population. This infection can develop into liver cirrhosis and liver cancer over the time in 80% of patients. In t...

Primary hepatocytes as targets for Hepatitis C virus replication

Much of our current understanding of hepatitis C virus (HCV) replication has hailed from the use of a small number of cloned viral genomes and transformed hepatoma cell lines. Recent evidence suggests that lipoproteins play a key role in the HCV life cycle and virus particles derived from the sera of infected patients exist in association with host lipoproteins. This report will review the lite...
